Alley, Milwaukee
Cal gets permission to take Skye on an outing from Afterlife to Milwaukee, where he used to have a medical practice. Gordon transports them into an alley.
The alley is called Frank Court and is located in downtown Los Angeles between 4th and 5th Streets. They are walking south with 4th Street behind them.

Sidewalk, Milwaukee
Emerging onto a sidewalk, Cal realizes things have changed a lot since he was here last. The walk along W 5th Street towards Spring Street, passing by a currency exchange store, which was added by the production team.

Ice Cream Store, Milwaukee
Later, Cal steps into a store to get ice cream while Skye steals a passerby’s phone to call for help. This takes them out onto approximately 420 S. Broadway, just outside the Placio De Oro, which is no longer there.

Interior Calvin’s Old Office, Milwaukee
Cal shows Skye his old medical office, which is where Bakshi tracks the SHIELD agents who have a showdown on the premises. This location appears to be the Spring Arts Tower at 453 S. Spring Street in Los Angeles. It’s an office building that allowed the production to shoot, apparently, anything they needed. This makes sense from a production standpoint since all the other Milwaukee scenes were filmed around this same block. The building was also used in the first season of Agent Carter.

Exterior Calvin’s Old Office, Milwaukee
A quick exterior shot of Cal’s office building shows the invisible Quinjet landing on the roof. This cityscape is not part of Milwaukee. It’s actually Vancouver, and the Quinjet appears to be landing on top of 900 W. Hastings St. The distinctive building with the stylized ‘H’ in the middle is the BC Hydro Building. Unfortunately, a new skyscraper between these two buildings blocks sight lines, no longer allowing for this view.
