Together these Sanctums generate a shield around the world.
DOCTOR STRANGE
The New York Sanctum is historically located at 177A Bleecker St, New York, as indicated by the placard on the exterior. However, the filming location for the Sanctum was done at 319 West 4th Street, on the 12th Street side of the building.
Strange steps out the W 12th St side of this building, looking East on 12th towards Greenwich Ave. Part of the exterior facade was a set piece while the rest of the front of the Sanctum is digitally created.

When the Sanctum shows up again in Avengers: Infinity War, it is located at it’s correct place in the city.

AVENGERS: INFINITY WAR
Stepping out of the New York Sanctum, Tony sees a giant spaceship hovering over New York.
While the scene appears to show a continuous shot of the characters exiting 177A Bleecker Street in New York and heading up Thompson St, the scene was actually filmed at the Atlanta Pinewood Studios, in Fayetteville, GA. Some clever set design and digital enhancement turned rural Georgia into downtown New York City. This video has some great behind-the-scenes footage of the set.

SPIDER-MAN: NO WAY HOME
Peter goes to get the help of Dr. Strange to create a spell to make the world forget that he’s Spider-Man. This leads to many unforeseen consequences, and turn Strange against Peter, who tries to do the right thing.
Due to the need to film lots of scenes in front of Dr. Strange’s Sanctum, this was not filmed in New York City, as with previous films depicting the Sorcerer Supreme’s domicile. The production only used background plates of the city and set in Atlanta.
A facade for the Sanctum was erected outside one of the stages at Trilith Studios (similar to the setup in Avengers: Infinity War), with everything beyond the immediate steps and doorframe being created digitally. This is evident in the street signs seen in the background. The street to Peter’s right, as he enters the Sanctum should be Sullivan Street, but in his first visit it’s labeled W 12th St, and later when he escapes with the magical box, W 4th St.
